N2 - It is 20 yr since the Ins(1,4,5)P3 3-kinase reaction was discovered, and this review summarizes and discusses some of the advances we have made in our understanding of the physiological significance of this reaction. The three major potential functions of Ins(1,4,5)P3 3-kinases are discussed in the context of their localization and regulation, the possible functions of their product, Ins(1,3,4,5)P4 and their potential contribution to the synthesis of higher inositol polyphosphates.
