Abstract
Transforming growth factor-β (TGF-β) controls the development and maintenance of skeleton. The mutations of TGFB and related genes lead to abnormal TGF-β signaling and various skeletal diseases. This subgroup of skeletal dysplasia can be divided as monogenic disease, which is caused by a single gene mutation, and polygenic diseases which is caused by combined effects of a group of genes as well as other risk factors. Studying these diseases enable us to understand mechanisms for development and maintenance of human skeletal tissue and lead to promising therapeutics for TGF-β-involved bone genetic diseases.
