Abstract
Self-nanoemulsifying drug delivery systems (SNEDDS) are known to possess immense potential for enhancing oral bioavailability of lipophilic drugs by facilitating the drug-release profile, inhibiting metabolism of CYP450, bypassing extensive hepatic first-pass effect, and potentiating absorption through intestinal lymphatic pathways. Although SNEDDS are effective in enhancing the systemic availability of the drugs, issues, such as poor drug loading, in situ stability, gastric irritation, and so on, are major concerns. In this regard, the functional excipients, including polar lipids, phospholipids, diblock polymers, are considered to be highly sought-after for developing specialized SNEDDS. Lately, modifications of SNEDDS into supersaturable, gastroretentive, and eutectic forms, have been used to address diverse related issues. Providing a comprehensive overview, the chapter endeavors to provide development and evaluation of variegated SNEDDS for oral bioavailability enhancement of diverse drugs.
