Abstract
Aim: To investigate whether PSD93 participates in the neurotoxity induced by platelet activating factor (PAF) so as to study the molecular mechanism of neuron injury due to PAF. Methods: The experiment was done in the Department of Neurology, Nanjing Drum Tower Hospital, and Dr. Tao Laboratory of the John Hopkins University from May 2003 to May 2004. PAF treated the cortical neurons of wild type and PSD93 knock out mice, and the cells were stained with propidium iodide/Calcein AM for apoptosis. Results: Neurotoxicity was obviously found in the cortical neurons of wild type and PSD93 knock out mice after treatment with 0.10, 0.30, 0.6 μmol/L PAP (P < 0.05). The apoptosis of cortical neurons in the PSD93 knock out mice was significantly lower than that in the wild type (P < 0.01). Conclusion: PSD93 knock out can protect cortical neurons from PAF-induced damage.
