Evaluation of isolated perfused segments of dog ileum as a means of growing tumor

Abstract

We were unable to maintain fragments of human tumor on the mucosal surface of isolated dog ileum as described by Folkman. With a radical modification of Folkman's technique we have been able to maintain HeLa cells injected submucosally. Modifications of the technique included a change in the perfusate, the gas mixture, and the oxygenator. Evidence of invasion by the HeLa cells was observed. On the basis of our present experience, the isolated dog ileum may be a satisfactory method of studying the behavior of various animal tumor systems, clones of human cells previously established in tissue culture, and perhaps fragments of biopsy specimens of human tumor.
