Abstract

There remains an unmet need for simple microfluidic platforms that can digitize and perform multi-step and multi-reagent biochemical assays in parallel for high-throughput detection and analysis of single molecules and single cells. In response, we report herein a vacuum-driven microfluidic chip that is capable of multi-step sample digitalization based on a tree-like multi-level bifurcation structure and gas permeability of PDMS. Multi-step loading capability and loading uniformity in each step were characterized, showing strong performance of our device. With further development, our platform can provide a promising tool for achieving high-throughput digital multi-step, multi-reagent biochemical assays.
